pBBR1MCS-2-TAC-EGFP Plasmid (2 µg) is a broad-host-range, low-copy-number, mobilizable plasmid engineered with a pBBR1 origin of replication, enabling replication across a diverse array of Gram-negative bacterial species, including Escherichia coli, Pseudomonas spp., and Agrobacterium tumefaciens. The plasmid carries a multiple cloning site (MCS) within the lacZα gene for blue/white screening and confers kanamycin resistance (Kan^R) for antibiotic selection. Downstream of the MCS, the tac promoter, a hybrid of the trp and lac promoters, drives constitutive or IPTG-inducible high-level expression of the enhanced green fluorescent protein (EGFP) gene, allowing for real-time, non-invasive fluorescence-based monitoring of gene expression and plasmid maintenance. The EGFP open reading frame is codon-optimized for bacterial expression and lacks any eukaryotic introns. This plasmid is ideal for fluorescence reporter assays, microbial tracking, and heterologous protein expression studies in environmental or synthetic biology contexts.
